mCherry is a deep red fluorescent protein that commonly used in the cellular localization studies of fusion proteins.
Single domain antibody (sdAb) is a 12-15 kDa sized antibody framgent of camelids. A sdAb against EGFP was screened using a fully synthetic sdAb gene library and purified by 6xHis affinity purification from E.coli after overexpression.
The GB1 domain from streptococcal protein G, binds to Fc region of IgGs, was genetically fused at the C-terminal end of sdAb protein. This enables an use of any commercial secondary antibodies for signal development in western and ELISA experiments.
